Yes.
It has already been told to not count towards the title.
Speculation is that the Lion that has been decaying will explode or collapse, opening up a new area to explore. So, up probably will not miss much, unless you really like exploding lions.

All you’re probably missing is a small scene, and potentially a short cosmetic event to announce the beginning of the end.
You are -not- missing Act 3. Just its opening.
