So should I remove my armor to be able to access it or unlock it when the update comes?
Or does it not matter and will just be placed in my locker when it is replaced by my PvE looks?

It doesn’t matter. Anything you are wearing will be unlocked for the Wardrobe. Anything in your PvP Locker will be unlocked for the Wardrobe and deleted.

You don’t have to do anything. Your inventory, equipment slots, locker, and account bank will all be cleared upon entering the mists. You’ll need to log into the mists with each character to get anything they are wearing also put in the wardrobe.
