My two concerns with HoT
Considering the new Precursor acquisition method (unlocked in HoT) sends you all over the game world to complete content, and that Specializations allow you to use the skills you unlock at any time, I would guess that you can use them anywhere.
This has a lot of interesting information about that: http://www.mmorpg.com/gamelist.cfm/game/473/feature/9340/page/2
In the presentation they emphasized the fact that they will not abandon the current game, and all aspects of GW2 will remain relevant.

with what I said I more mean masteries. how it was talked about made it seem like it’s exclusively for maguma.

Well it does sound like you will be able to get Mastery Points all across the game-world so it is rather likely that the Masteries can also be used everywhere.

Well, they say here that Masteries will be used all through the game, including crafting, which (I assume) won’t be Maguuma Jungle-exclusive…
The WvW ability system is a bit of a parallel for what we’re trying to accomplish with masteries, so you can use that as a concept. We want to provide that same concept and evolve it even more with PvE. It’ll complement everything that you’re doing: combat, exploration, lore, building, crafting… all of those components will have elements that are supported by it, with combat being the most important one when you think about true, meaningful progression.
It may be that you can only earn Mastery points in Maguuma….I don’t know. I guess we will find out when they start doing the ‘Deep Dives’! =)

I don’t think that is true actually. I think the interview in which Mike said that is misunderstood. I understood more like they would add Mastery points to all of Tyria later on, not now.
Otherwise, Mike and Colin said that some of the encounters in the Maguuma can’t be finished without having the correct Masteries so I think they are far more area/event-specific abilities than kitten skills that are just added to your character.
