I have a couple friends who have started playing the free-to-play version bu they want to have access to the full core game. If they pre purchase HoT will the core game unlock for them now or do they have to wait until HoT release to gain the core game benefits?

Yes, yes it does unlock it at the moment they register the key.

You can only purchase Heart of Thorns now, at least through the official Shop, which includes the core game. It should unlock the core game experience instantly, and Heart of Thorns as it releases on October 23rd.

You can purchase though Gamestop too. That what I did, but I was already a B2P account for 3 years now.
