From my understanding you unlock tracks with mastery points and then progress along those tracks by leveling.
CureForLiving is correct, yon train the mastery tracks with exp, and you get mastery points from various content such as personal story, adventures, and mastery commune orbs.
I was in the stress test and there were 2 Mastery Point commune spots, like skill points, and I got 2 more from doing content in there.
So, as far as I understand it, it’s like this (MP=mastery point. XP=experience)
[MP] —-— XP —-—> [MP] —-—- XP —-—> [MP]
You gain MPs from doing content and when the XP you have gotten reaches an MP slot (the mastery line is “trained”), you can put the MP in that MP slot.
ANet may give it to you.
From what I understand:
“level up” past 80 will work much the same as now, with one exception:
Mastery point instead of skill point.
How it will determine which regional mastery point ( eg hot vs vanilla) isn’t known. I suspect it may be determined by the region you are in when you level up).
You can also slot a mastery line in place of your old exp bar.
Then experience you gain will progress(train) that mastery line.
Each mastery line will use X exp to train. N will likely vary from mastery to mastery, and from tier to tier of the same mastery.
Example tier one of gliding may require 1 mastery point, then 254k exp to train.
Tier 2 of gliding may require 2 mastery points, and then 508k exp (there may not be a direct correlation between number of mastery points and exp points required to train however).
I expect exp granting consumables will still grant the same amount of experience points they do now ( in the case of ToK, 100% required for current level up, or 254k at 80).

Commenting on: a couple of things you said

You said, can replace. They said that the new mastery line will replace the old exp line after level 80. It won’t be optional, and your level 80 char will no longer show 80 as a character level but will now show how many Mastery Points your account has gained. (You do have to chose which one you want to be currently training. Perhaps that’s what you meant).
And: “in the case of ToK, 100% required for current level up, or 254k at 80”
One guy got a Tok during the stress test in a container. He used it and only got one-fifth of the mastery bar’s progression. So it appears at best a Tok will give only a fraction of a Mastery bar increase.

I’m just worried that 1 character won’t be able to attain all mastery points; that you have to choose certain mastery lines you can follow but that each character may only do enough content to max out 1, perhaps 2 masteries. Since these are account-wide, it would only make sense if this was the case, however that would force people to play other characters to max out all masteries..
Ex: I max out exploration on my warrior due to thinking it will unlock more content only it could (ie with the hang glider skill). It does, however I next go in to combat mastery, and do all content available. It only fills my mastery bar half way…now I have to log in to my ranger at level 35, level him up, and continue earning mastery points to fullfill my combat mastery bar.
Of course, each mastery may unlock potentially more options to earn points, and as this is all simply speculation until the expansion hits, I state this only as opinion; i hope wel dont have to log in to multiple characters to max our masteries.
Just a quick question/speculation.
As I understand you get 1 mastery point for each lvl up after lvl 80(in HoT)These scrolls you get for dailies, are they gonna give you 5% of a mastery point?
Does ToK gonna get you mastery points?
The skill scrolls still give regular skill points I guess?

Leveling won’t give you any mastery point. Mastery point are unlock through success/living story/special node in game. You’ll then use these mastery point to unlock part of mastery track that need to be filled with experience to obtain their effect.
Skill scrolls will still give skill point.
Tome of knowledge will give experience. (There have been testing in the Beta, 1 tome of knowledge fill around 1/5th of the first stage of a mastery track).
Experience scrolls will then probably end up giving 1% of the experience needed to finish 1 stage of a mastery track.
