I’m not exactly sure what you’re asking for. You can either hold ctrl and click the target, which seems like what you’re doing. If you already have the mob targeted, you can just hit ctrl+t (not sure if that’s default or not) to mark it, without clicking again.
I have a mouse button mapped to Mark the target and another one mapped to Go To the Marked target. I just target the mob I want to mark (either click on them or tab target) and click my Mark Them mouse button and the target is marked. And if I lose the target, my Go To Marked button puts me right back on target.
I have never had to hold down any keys to mark the target. What ever mob I have targeted when I hit the hot key or use my mapped mouse button get the mark. But I do always start the process by having a target selected before I press the hot key.
I only wish ArenaNet would allow us to use the Mark and Go To Marked when solo. It would certainly give some relief to the accidental right-click untargeting issue.
Does anyone know how to remove the target mark once the mob is dead? I usually just mark one of my guildie teammates and let them run around with it on until we find the next big mob to kill. Kinda funny but I’d really love to know how to do this without messing with my teammates.

I see, select the target by clicking them and then hit the bound button to mark them… yes I was doing it by holding ctrl and clicking them. Thanks!
