I viewed the new interface for modifying your build in Heart of the Mists, and noticed that there is only 1 specific spot for your rune. Does this mean you are removing the mix and matching with the runes? (i.e. having a 2-piece bonus from one rune, 2-piece bonus from another rune, and a third 2-piece bonus from another rune)

Yes, there is no more mixing and matching (in pvp) – you take all 6 benefits of 1 rune-set.

In PvE/WvW, you can still mix and match, but the benefit of doing so has been greatly reduced. More or less, there is strong incentive to invest in a full set of 6 runes.
