Yes they do, they are considered shouts, I -believe- it removes one from you -and- your pet (friend who uses the tune swears it removes from everyone affected), but idk about that.
I -do- know it’ll remove a condi off you.

Yes, the rune 6 skill does trigger when you use ranger shouts. But you are better off using a warrior with rune of the warrior, since their shouts have lower recharge.
